When analyzing data, sometimes we feel very frustrated because just as one problem has been addressed, there is always another one just around the corner. It feels like it never ends, especially since we still need to meet each frustration and deal with it. When modeling, we need to pay attention to common problems, like those involving missing values, outliers, limits of detection, and unusual distributions, which can make all the difference between drawing the right conclusion – or the wrong one.

How to Make More from Online and Offline Fermentation Data to Speed up Process Development

Resources in process development for fermentation processes are always limited, often because of time constraints or the limited access to fermentation vessels. For this reason, the optimal usage of the data collected is of utmost importance.
This presentation shows how to maximise insights and understanding of the process with by modelling data collected offline (like product titer and parameters of downstream processing) and online sensor data (like pO2) in context of the Functional Data Explorer platform. The modelling approach is based on constant and functional factors and responses. Because of the limited design space, the new extrapolation control for the profiler plays an important role here.
Furthermore, this presentation highlights the advantages of using Graph Builder to lead a team and subject matter experts to a faster, easier and more efficient understanding of the produced data so that they may make better and more reliable decisions in the fields of bioprocess development.

MSA and Optimization of a UHPLC Measurement System

An ultra high performance liquid chromatography (UHPLC) measurement system analysis (MSA) optimization and validation study in a quality lab is presented. Process settings of the analysis method are established in order to maximize measurement accuracy and resolution of two organic compounds. There are seven control factors and optimal DOE is used to specify how the experiments take into account specified model and experimental criteria. It demonstrates why OFAT is not appropriate and how to decide between a custom DOE and a DSD based on DOE diagnostics such as power, effect correlation and variance profiles. Using stepwise regression, good predictive models are obtained that are supported by validation experiments. The Profiler desirability function is used to determine the optimal and robust UHPLC settings for measuring both compounds. The particular importance of the sensitivity indicator for improving robustness is shown.

Predicting Molecule Stability in Biopharmaceutical Products with a Rapid Short-Term Study Using JMP

In the journey of delivering new medicines to patients, the new molecular entity must demonstrate that it is safe, efficacious, and stable over prolonged periods of time under storage. Long-term stability studies are designed to gather data (potentially up to 60 months) to accurately predict molecule stability.
Experimentally, long-term stability studies are time consuming and resource-intensive, affecting the timelines of new medicines progressing through the development pipeline. In the small molecule space, high-temperature accelerated stability studies have been designed to accurately predict long-term stability within a shorter time frame. This approach has gained popularity in the industry but its adoption within the large molecule space, such as monoclonal antibodies (mAbs), remains in its infancy.
To enable scientists to design, plan, and execute accelerated stability models (ASM) employing design of experiments for biopharmaceutical products, a JMP add-in has been developed. It permits users to predict mAb stability in shorter experimental studies (in two to four weeks) using the prediction profiler and bootstrapping techniques based on improved kinetic models. At GSK, Biopharm Process Research (BPR) deploys ASM for its early development formulation and purification stability studies.

Comparing Predictive Model Performance with Confidence Curves

Repeated k-fold cross-validation is commonly used to evaluate the performance of predictive models. The problem is, how do you know if a statistical fluke makes one classifier appear better than another? Typically, null hypothesis significance testing (NHST) is used to determine if the differences between predictive models are “significant.” In this presentation, we discuss problems associated with NHST and present an alternative known as confidence curves, which has been developed as a new JMP add-in that operates directly on the results generated from JMP Pro's Model Screening platform.

Finding the Optimal Parameters for Laser Welding of Steel Plates with JMP

As a supplier of mechatronic solutions for the steel industry, Clecim recently extended its range of laser welders by offering a new generation of machines capable of cutting and welding heavy plates using a 12kW solid-state laser.
To be considered good, a weld bead must meet two criteria. It must be free of defects (such as spatters, humpings, underfilling, holes, etc.) and it must be resistant (evaluated by means of an Erichsen-type cupping test).
To achieve this result, many steps using JMP features and platforms were implemented. The first was determining the base material strength and characterization of the two plates to be welded, specifically the average strength and strength profile.
Next, visual observations were synthesized and the welding defects map was obtained, according to the studied parameters, to determine an irregularly shaped study area where the weld bead was defect-free.
Finally, the weld bead strength on a non-homogeneous material and the previously determined defect-free zone were analyzed and optimized, using a set of candidate points.

Biodiversity loss is a global challenge. Reliable data on the numbers and distribution of species are urgently needed to stem the species hemorrhage. But getting those data is hard. Endangered species are elusive and cryptic, and current monitoring techniques often expensive and unreliable.
Frederick Kistner is a founder member of the WildTrack Specialist Group. Together with colleagues Larissa Slaney from Heriot-Watt University, and Zoe Jewell and Sky Alibhai from WildTrack, he is developing a method to identify four overlapping species of otter that exist in southeast Asia: the Eurasian otter (Lutra lutra), the short-clawed otter (Onyx cinereus), the smooth-coated otter (Lutrogale perspicillata), and the hairy-nosed otter (Lutra sumatrana).
Morphometric features of otter footprints are extracted using a customized JMP script, implemented in the Footprint Identification Technology (FIT) add-in. The data are partitioned using the JMP validation column creator. Morphometric features are used as input variables to predict the otter species as the target variable. The automatic model selection feature in JMP then identifies the best model. Initial findings comparing prints from Asian short-clawed otters with Eurasian otter yielded 100% classification accuracy on the training (50%), as well as on the test set (50%).

Biopharmaceuticals' New Frontier: Modeling Patient/Donor Variability for Gene/Cell Therapy in JMP

The most novel, innovative and promising therapeutics in biopharmaceuticals are gene and cell therapies. Gene therapies modify a patient’s existing genes, while cell therapies transfer human cells into a patient to treat disease. These cells either come from the patient themselves or a healthy (cell) donor. The first such therapy approved in the United States was Kymriah in 2017. Since that time, there have been countless companies exploring these novel approaches to treating patients. One of the main barriers to regulatory approval is the development of a process that consistently meets the needs of patients (in both efficacy and safety). One obvious challenge is modeling either patient-to-patient or donor-to-donor variability that can (and usually does) account for a large portion of the variability.
This talk focuses on different methods of modeling donor-to-donor variability in JMP to answer important research questions: determining significant process parameters (independent of patients/donors), constructing proven acceptable and normal operating ranges (PAR and NOR), and estimating process capability. After demonstrating the implications of modeling donor as a fixed block effect, the authors then present the advantages to modeling donor as a random effect, first by presenting modeling and simulation features in JMP and then additional features in JMP Pro.

Controlling Extrapolation in the Prediction Profiler in JMP Pro 16

Uncontrolled model extrapolation leads to two serious kinds of errors: (1) the model may be completely invalid far from the data, and (2) the combinations of variable values may not be physically realizable. Using the Profiler to optimize models that are fit to observational data can lead to extrapolated solutions that are of no practical use without any warning. JMP Pro 16 introduces extrapolation control into many predictive modeling platforms and the Profiler platform itself. This new feature in the Prediction Profiler alerts the user to possible extrapolation or completely avoids drawing extrapolated points where the model may not be valid. Additionally, the user can perform optimization over a constrained region that avoids extrapolation. In this presentation, we discuss the motivation and usefulness of extrapolation control, demonstrate how it can be easily used in JMP, and describe details of our methods.

When you collect data from measurements over time or other dimensions, you might want to focus on the shape of the data. Examples can be dissolution profiles of drug tablets or distribution of measurement from sensors. Functional data analysis and regression-based models are alternative options for analyzing such data. Regression models can be nonlinear or multivariate or both. This presentation compares various approaches, emphasizing pros and cons and also offering the option to combine them. The underlying framework supporting this work is information quality, which permits us to consider the level of information quality provided by the two approaches and the possible advantages in combining them. Implementation of a Split-Plot Design Platform to Study the Purification of Innovative Excipients

The BioChaperone platform developed by Adocia covers oligomers of various sizes. However, they have the common characteristic of being of higher molecular weight than the impurities generated during their synthesis process. Thus, their purification can be achieved by diafiltration or tangential filtration. This technique, thanks to a membrane with pores of defined size, makes it possible to separate the molecules according to their molecular weight. Different parameters such as temperature, tangential flow rate, and pressures have impacts on the purification efficiency, as well as on the duration of the operation. However, the multivariate study of this step was hampered by technical constraints, such as the impossibility to change the temperature between each trial, thus limiting the randomization. Therefore, a split-plot design approach implementing a randomized block system was developed to optimize the purifications of our excipients.

Importing Binary Data with JSL

JMP provides many data importing methods, allowing you to retrieve data from large databases down to simple text files. But what if your data is in a unique file format that JMP cannot directly import? By using the powerful but little-known JSL Blob (Binary Large OBject), you can extract that valuable data for use in your analysis.
This presentation offers a case study in developing an import function for a common but obscure data source: the metadata locked within a digital photograph. Modern cameras record much more data than just the image.Metadata -- how, when, and where the photo was taken -- is locked away within its binary EXIF tags; with JSL Blobs, we can import that data into JMP. From there, we can use JMP's analytic tools to measure metrics of our photo library, and even produce a map-based display of our photos.

SVEM (Self-Validated Ensemble Modeling): A Path Toward DOEs with Complex Models and Very Few Runs

In industrial development, especially if experiments are conducted on production scale, the number of DOE runs required to cover the actual problem is always too high, either in terms of costs or time. At the 2021 Discovery Summit, the introduction of SVEM (self-validated ensemble modeling) caught my attention, due to its power to build accurate and complex models with a limited number of experiments. Especially in the area of costly experiments, SVEM opens a way to fit complex models in DOEs with considerably reduced amount of runs, compared to classical designs.
Three case studies are presented. The first two case studies deal with designs conducted on a production scale, first on a five-factor, 13-run design and then on a four-factor, 10-run design, each with nonlinear problems. Another use case shows a Bayesian I-optimal 15-factor, 23-run design for a nonlinear problem. Especially within the first use case, the excellent predictive accuracy of the models obtained by SVEM led to the discovery of faulty measurement equipment, as measurement results started to deviate from the predicted results. I'm convinced that SVEM has the potential to effectively change the way DOE will be applied in product development.

Textual analysis of written documents has become an important analytics tool in accounting and finance decision making. Several research papers have expanded the textual analysis and have also measured the written tone in financial documents converting the tone a quantitative score of optimism/pessimism. Some of the research has connected the tone to explain abnormal returns in the financial markets. We explore this connection with the use of the Sentiment Analysis platform in JMP Pro 16.

One Multi-tabbed, Filterable Dashboard

Data collection teams within companies amass amazingly large amounts of information. And while these data are very valuable, it is not often tranlated into a format that is usuable by the recipient. Further compounding the problem is that different business units consume the information in different ways, which, unfortunately, often meants that valuable information is never consumed or utilized.
How does one best translate the needed information into an easy-to-use format? Furthermore, can statistically significant information be communicated through one interface? This paper describles the creation of one multi-tabbed, filterable dashboard to communicate computer laptop systems data to both architecture and engineering organizations.

Optimizing Multivariate Models: From Data Collection to Optimal Conditions and Scoring Models

Manufacturing processes are highly correlated systems that are difficult to optimize. This correlation provides an opportunity to identify outliers. Here, we demonstrate an entire workflow, which involves:

Obtaining data from the OSISoft historian.
Modeling and optimizing with multivariate models that account for process constraints.
Using the MDMCC platform.
Scoring models online and writing abnormal conditions back to the historian.

In practice, many processes are hard to optimize. Sometimes a system cannot be reduced to a set of independent variables that can be tested; in other cases, the process can become unstable under different conditions. To address these issues, we are using new features in JMP 16 to optimize for cost and quality in the latent variable space, while accounting for constraints on observed variables. In order to remain at those optimal conditions, we use the MDMCC platform to quickly identify deviations from normal.
Add-ins are used to minimize the time spent gathering data and scoring models: one to pull process data into JMP from the popular PI historian, one to quickly recreate calculated columns, and a third to score models online and send results to the historian, including contributions from the MDMCC.

Damien Bonnet, Validation Engineer, Thermo Fisher Scientific

For the past four years, Damien has served as Validation Engineer and Continued Process Verification Leader at the Patheon Bourgoin site outside of Lyon, France. With 6 years of experience in the pharmaceutical and medical device industries, Damien has worked primarily on risk assessment, qualification and validation activities, including at both Sanofi Pasteur (vaccines) and Allergan (syringes), before turning completely to the process monitoring of solid forms.

Véronique Brianto-Gabier is a Validation Engineer and Continued Process Verification Deputy for the Pharma Services Group of Patheon, a Thermo Fisher Scientific company. Before joining Thermo Fisher Scientific in early 2020, Brianto-Gabier was Head of Project Industrialization and Process Characterization for Biom'Up, a maker of collagen-based medical devices for biosurgery. A Lean Six Sigma Green Belt, she holds a Master’s in Physiology and Neuroscience.

Owen Lozman, Managing Director, M Ventures

Dr. Owen Lozman, FRSC CChem CSci, is Managing Director at M Ventures and head of technology investments. The M Ventures Technology fund invests across a broad range of topics with strategic relevance to the Electronics Business of Merck (KGaA, Darmstadt Germany), as well as covering cross-sector topics relating to digitalization and sustainability. Previously, Lozman worked for the parent company Merck (KgaA, Darmstadt, Germany), where he was head of New Platform R&D for the Electronics business. He was also responsible for leading a world-class global team of scientists, including the Electronics R&D centre, based in Chilworth, Southampton, UK. During his career in Research and Development, Lozman has driven many successful projects from concept to commercialization, working closely with customers and partners to bring disruptive technologies to market, as well as securing increased profitability for mature product lines. Lozman has previously worked with FujiFilm, Avecia and the University of Leeds. He holds a PhD in physical organic chemistry from the University of Leeds (UK) and has published more than 50 papers and has 15 patents.

A Lasso Regression to Detect Risk Factors for Fatal Outcomes in Critically Ill COVID-19 Patients

After nearly two years, our experience during the COVID-19 pandemic has made us all experts in knowing the risk factors for developing a severe case. For example: male, advanced age, obese, hypertensive -- right? Well, it depends. When we analyzed data from the subgroup of the most endangered patients -- those who were already hospitalized and in critical condition -- we discovered some surprising differences with respect to the common risk factors.
With a binary response (recovered/dead), fitting a logistic regression model seemed to be a reasonable approach. Due to the high dimensionality of the data, we used a penalized (Lasso) regression to select the most relevant risk factors. In our talk, we briefly introduce penalized regression techniques in JMP and present our results for critically ill COVID-19 patients.

Control Charting Kinetical and Other Time-Dependent Curves

SPC and control charting is a common procedure in industry. Normally, you are controlling and observing a single measure over time. These data are displayed with ± 3s limits around the mean on a chart.
However, when kinetical curves or other time-dependent behaviors are a matter of quality and consistency of a process, it is much more difficult to display in a SPC chart. These curves are often displayed within their maximal and minimal specification for each time point, which makes the off-spec curves visible. How can “off-spec” curves be defined while staying within these max-min limits?
The first method to try might be the principal component analysis (PCA). If the runtime stamps are always the intervals, then it's easy to achieve results. However, if they vary, interpolation of the Ys that will be on the same stamp is needed, which complicates data preparation.
With the Functional Data Explorer in JMP Pro, it becomes very convenient to display the different curves as principal components in a T 2 -control chart.
This presentation shows how we used this tool for quality control for a pressure leakage test and how we made it simple for the practitioner to use.

OMARS Designs Add-in: A Gateway to a New Family of Orthogonal RSDs

The definitive screening design (DSD) is almost certainly the 21st century's most exciting and useful innovation in design of experiments (DOE). As a screening design, the DSD offers unique properties for a much smaller number of a runs. And, if only a half or fewer of the factors are active, the DSD gives you the ability to fit the full response surface model. However, if your objective is to estimate the full response surface model for most or all of your factors, a DSD is inappropriate. In those instances, larger optimal designs or central composite designs (CCDs) are the preferred choices. Orthogonal minimally aliased response surface (OMARS) designs are a new family of response surface design (RSD) that bridges the gap between the small, efficient DSD and the large, high-powered CCD.
In this presentation, we introduce OMARS designs by way of a case study comparison with other designs. We also demonstrate how JMP users can create and evaluate OMARS designs against DSDs and classical RSDs in an easy-to-use add-in that will help you to select the right design for your specific application.

Choice Design and Max Difference Design/Analysis in Optimizing High School STEAMS Projects

This presentation demonstrates how consumer research methods, choice and max difference design modeling and analysis platforms in JMP 16 were used to help high school students optimize their utility and satisfaction in purchasing the right laptop for school usage, pick the right school courses, maximize their exam testing performance, and optimize their time spent on high school STEM projects (with a STEAMS approach).
For example, Choice Design and Model platforms were used to conduct survey analysis of laptop purchasing preferences. MaxDiff Design and Model platforms were used to conduct survey analysis of the popularity and difficulty of school courses.The Item Analysis platform was used to study exam question profiles to help students and instructors assess exam difficulty. The Latent Class Analysis platform was used to study multiple choice exams. Finally, using Explore Patterns/Explore Outliers, potentially unusual patterns in responses among examinees were detected, thus uncovering possible evidence of exam cheating. This work demonstrates how modern choice design can be used to optimize survey methodology that avoids sampling bias and how to use choice model to analyze survey data.

Defect Subpopulation Models

Suppose a small proportion of a population of components is highly susceptible to a certain failure mechanism. Consider these units to be manufacturing defects in a reliability sense: working originally and not detectable as damaged by standard end-of-the-line tests and inspections, yet after a short period of use, they fail. The rest of the components in the population are either not susceptible to this failure mechanism, or else they fail much later in time for possibly some other cause.
Occasionally, field or life test data indicates this bimodal behavior by showing an early incidence of failures that seem to slow down to almost nothing long before a significant proportion of the population has failed. A good model for this kind of behavior is the defective subpopulation model (DS).
The population of components is really a mixture of two populations: a reliability defective small subpopulation that fails early and the remaining components that may possibly fail later due to wear-out mechanisms that only are seen much further out in time (designated as “immortals”). This model, where a (hopefully small!) proportion of the population follows an early life failure model, has wide applicability.
This talk first describes how such data can be analyzed using the Life Distribution platform in JMP through the DS options for fitting the data and then demonstrates how to apply a simple test of the hypothesis that there may be a defective subpopulation based on the output of the Life Distribution platform.

Well, if you happen to have an interest in aquaculture and a background in chemistry, they have the makings of a decent spectrometer. If you throw JMP into the mix, then you have the makings of a system that can be used to detect the presence of harmful waste byproducts in a fish tank with startling accuracy. Join Mike Anderson on his adventure in setting up and “cycling” a new 36-gallon fish tank using a homemade spectrometer and JMP Pro’s Functional Data Explorer to monitor the process.

47 Years (So Far) with Designed Experiments from One-Factor-at-a-Time Designs to GO-SSD Designs

Like most young engineers at Kodak, I started out running one-factor-at-a-time designs, until we needed to optimize a five-component solution. After successfully optimizing this solution that had two statistically significant three-way interactions, I was hooked on designed experiments. As computer tools progressed, we moved from pencil and paper to in-house mainframe programs. In the 1980s we started using mainframe SAS. In the 1990s we had JMP, but our DOEs were still basically catalog designs. In the late 1990s when JMP added optimal designs, we had some new tools. Experiments didn’t have to be orthogonal. We could run smaller experiments and still get the necessary data, which was very important in the waning days of the Kodak Research Labs when strict quotas were placed on the number and size of experiments. When Bradley Jones and Christopher Nachtsheim introduced definitive screening experiments in 2011, I had moved on to Bausch + Lomb. We have had more than one case where DSDs identified antagonistic interactions early enough to avoid program delays. DOEs in retirement are simpler, focusing on things like optimizing an ice cream recipe and reducing the amount of time to produce maple syrup on a conventional stove.

A JMP Script That Determines a Simultaneous 95% Bound Using a K-Nearest Neighbor Approach

We have a set of responses that follow some continuous, unknown distribution with responses that are most likely not independent. We want to determine the simultaneous 95% upper or lower bound for each response. As an example, we may want the lower y1 and y2 and upper y3 and y4 bounds such that 95% of the data is simultaneously above y1 and y2 and below y3 and y4. Finding the 95% bound for each response leads to inaccurate coverage. The solution: a method to calculate the simultaneous 95% upper or lower bound for each response using nearest neighbor principles by writing a JMP script to perform the calculations.

A Surprising Use of the Fit Definitive Screening Platform

As is evident by its name, the original intended use of the Fit Definitive Screening platform was to analyze Definitive Screening Designs (DSDs). The surprise is that this platform can analyze a much broader class of designs than just DSDs. It turns out DSDs are a very special kind of Foldover Design, which are a standard textbook design used for factor screening. All that's needed for Fit Definitive Screening platform to do its innovative analysis is a Foldover Design. This talk demonstrates how to make Foldover Designs using the Custom Design tool and then analyze them using the Fit Definitive Screening platform. Several examples illustrate this two-step procedure and the analytical results are compared with more standard approaches that ignore the structure of the design.
